Transfer Plasmid
Drug Discovery & Development - August 01, 2010

Oxford Expression Technologies, a specialist provider of baculovirus-based protein expression solutions, launches pOET3, a novel baculovirus transfer plasmid for improved expression of glycosylated, secreted, and membrane-targeted proteins.

pOET3 combines the flexibility and convenience offered by the pOET1 and pOET2 vectors, with enhanced capabilities that complement the improvements in protein expression delivered by the Oxford’s flashBAC vectors, the company reports. pOET1 and pOET2 are baculovirus transfer plasmids designed for high-level expression of foreign genes under the powerful AcMNPV polyhedrin (polh) promoter.

pOET3 protein expression is driven from the AcMNPV p6.9 promoter, enabling earlier expression compared with the polh promoter used in pOET1 and pOET2. The recombinant protein is therefore produced at a time during the infection process when the cellular machinery is in a much healthier condition, compared to late times in infection. This can give significant improvements when proteins that require extensive post-translational modifications are produced, such as glycoproteins destined for secretion or membrane insertion.
